
Domainex will build a pipeline of small-molecule drugs targeting enzymes that utilise nucleotide triphosphates (NTP) as substrates or cofactors. Many of NTP-dependent enzymes are well-validated therapeutic targets, and nucleotide binding pockets are recognised to be highly druggable sites. However, many of these exciting drug targets are intractable using conventional protein expression methods, and biochemical activity is frequently only seen with the full-length protein.
Domainex is therefore developing a combination of approaches, which we call DomaiNTaP, to address these issues. Our Combinatorial Domain Hunting technology can be used to deliver stable, soluble, folded and highly expressed protein constructs; generic affinity-based screens will allow us to identify nucleotide-binding constructs and chemical hits rapidly; and the LeadBuilder platform will afford cost-effective translation to high-quality drug ‘leads’.
